In the older group, NR raised NAD(P)H, lowered a marker of oxidative stress by about 18%, improved isometric peak torque by 8%, and improved the fatigue index by 15%. In the young group, it did none of that. The authors' interpretation: redox supplementation helps mainly when there's an underlying deficiency to correct. Older muscle starts lower; younger muscle doesn't have the same headroom.
Does pairing NAD+ with training help?
Here the evidence gets messier and more honest. Reviews of NR and exercise note that combining NAD+ precursors with training has produced conflicting results, with some animal work even suggesting NR could blunt certain exercise adaptations by interfering with redox signalling that exercise normally uses as a positive stress. That's a genuine note of caution, not a selling point: more NAD+ is not automatically better for a body that's adapting to training.
Other human work supports NAD+'s role in muscle more broadly — NR augmented the NAD+ metabolome in aged skeletal muscle and shifted anti-inflammatory signatures — but "changes muscle biochemistry" is several steps short of "makes you fitter or faster."

Why age seems to matter so much here
The single most striking feature of the exercise-and-NAD+ literature is how often the effect shows up in older participants and disappears in younger ones. In Dolopikou's acute trial, the improvements in isometric torque and fatigue resistance, and the drop in oxidative stress, were seen in the older group — not in the young adults given the same protocol. The most plausible reading is that younger people generally aren't NAD+-limited for exercise, so topping up a system that isn't short doesn't add much. This is a useful filter for your own expectations: a fit 25-year-old and a 65-year-old are not the same candidate for any performance effect.
What this does and doesn't justify
None of this supports the idea that an NAD+ precursor is an ergogenic aid in the way creatine or caffeine have earned that label through repeated trials. The exercise signals are small, somewhat age-dependent, and not consistently replicated across endpoints.
