How does caffeine actually keep you awake?

Through the day, a molecule called adenosine accumulates in the brain and builds what sleep scientists call sleep pressure — the growing drive to sleep. What human studies actually show

A 2025 placebo-controlled, double-blind crossover trial (Stone and colleagues, published in Sleep) tested dose and timing directly. Twenty-three participants took either 100 mg or 400 mg of caffeine 4, 8, or 12 hours before bed, with in-home partial polysomnography. The 100 mg dose produced no significant effect on sleep at any of those timings. The 400 mg dose was a different story: it delayed sleep onset and altered sleep architecture even when taken 12 hours before bed, and reduced perceived sleep quality by about a third when taken 4 hours before.

An earlier classic (Drake and colleagues, 2013, Journal of Clinical Sleep Medicine) gave 400 mg at 0, 3, or 6 hours before bed and found significant sleep disruption at all three timings versus placebo — including the 6-hour mark, which is where the common "cut caffeine 6 hours before bed" rule comes from.

So what's a realistic cutoff?

The pattern across trials suggests a moderate single dose (around 100 mg, roughly one regular coffee) is reasonable up to about 4 hours before bed for most healthy sleepers. Large or stacked doses — a strong pre-workout, energy drinks, several coffees — deserve a much wider buffer, on the order of 8 to 12 hours, because the evidence shows they can reach into the night even from the morning. What caffeine timing won't fix

Cutting caffeine earlier helps, but it won't resolve insomnia driven by stress, an irregular schedule, or an undiagnosed condition. If you've moved your cutoff to mid-morning and still can't sleep, the caffeine probably wasn't the main lever — look at consistency, light exposure, and the broader basics of better sleep.

Why "it doesn't affect me" is so unreliable

Genetics explain a large part of why people differ in caffeine sensitivity. Variation in the CYP1A2 gene determines how fast the liver clears caffeine: fast metabolizers process it in a few hours, while slow metabolizers can have meaningful amounts circulating well past midnight from an afternoon cup. A second gene, ADORA2A, shapes how strongly the adenosine receptor responds, which is why two people with the same coffee habit can have completely different nights.

The deeper problem is perception. In controlled trials, caffeine measurably reduces deep sleep and total sleep time even in participants who report no difference at all the next morning. The alerting effect masks the cost, so self-assessment systematically underestimates the damage. If you suspect caffeine but "don't feel it," the more reliable test is to cut your last dose earlier for two weeks and watch whether you stop waking in the small hours, rather than trusting how alert you feel.

Frequently asked questions

"Caffeine doesn't affect my sleep." Is that possible?
Some people metabolize caffeine faster genetically, so there's real variation. But the trials are humbling: people frequently slept objectively worse without noticing, so self-report is shaky evidence here.

Does decaf count?
Decaf still contains small amounts of caffeine, but typically far below the threshold that disrupted sleep in these trials. For most people it's not a meaningful concern late in the day.

What about tea or matcha?
It's the total caffeine dose and timing that matter, not the source. A strong matcha or large tea can carry a meaningful dose; a light cup usually doesn't.
